Corneal lipidosis in rats treated with amphiphilic cationic drugs.

The purpose of this study was to investigate whether generalized lipidosis experimentally induced by cationic amphiphilic drugs in rats is regularly associated with lipidotic alterations in the cornea. After chronic oral treatment with chlorphentermine, iprindole, or tamoxifen all animals showed clear lipidosis-like alterations in corneal cells. After chronic oral treatment with chloroquine and quinacrine the results were variable and unpredictable; however, consistent lipidosis-like alterations were found when chloroquine was applied locally onto the cornea. The present results show that basically the cornea of rats is involved in drug-induced lipidosis. For toxicological studies it must be kept in mind, however, that the reactions of rat cornea may be unreliable and less marked than in the cornea of human beings.

Effect of chlorphentermine pretreatment on the distribution of chlorphentermine in isolated perfused rat lungs.

Previous studies have shown that a 7-day treatment of rats with chlorphentermine (CP) enhances total pulmonary phospholipids and significantly enhances the accumulation of CP in perfused lungs. This study was conducted to determine if the accumulation of CP was associated with a particular fraction of the lung, and if the enhanced uptake correlated with the increased phospholipids. Rats were treated daily for 7 days with 50 mg/kg CP dissolved in saline and pair-fed controls received the vehicle only. On Day 8, the rats were sacrificed, and the lungs were removed, ventilated, and perfused. [14C]Chlorphentermine (5 mumol) was added to the perfusate of control and CP-pretreated lungs. After perfusion, the lung was homogenized and subjected to standard fractionation procedures. Some lungs were examined by light and electron microscopy. After 60 min of perfusion, CP uptake by lungs obtained from CP-treated rats was enhanced in comparison to uptake by lungs from control rats. However, CP was distributed uniformly among subcellular fractions, and CP pretreatment did not alter this pattern. Pulmonary macrophages obtained from CP-treated animals contained 8 times more CP than controls. Increased CP uptake following pretreatment can be accounted for by increased CP in the macrophages of treated rats. Macrophages in the lung tissue and in the lavage fluid from rats treated with CP were more numerous and larger than in controls lungs. This suggests that a close association may be found between accumulated CP and macrophage uptake.

Amphetamine-induced hypodipsia and its implications for conditioned taste aversion in rats.

According to the conditioned anorexia hypothesis, conditioned taste aversions occur when flavour stimuli are classically conditioned to the anorexigenic or hypodipsic effects of drugs. The effects on water intake of a range of doses of amphetamine and of several related compounds have therefore been examined in an attempt to correlate their known potentices in tate aversion experiments with their hypodipsic potencies (+)-Amphetamine was more potent than (-)-amphetamine in suppressing water intake but under similar experimental conditions, the isomers were equipotent in the conditioning of taste aversions. Methamphetamine and p-chloromethamphetamine were equipotent in suppressing water intake, but the latter was a more potent agent for conditioning taste aversions. Furthermore, fenfluramine produced taste aversions at doses well below those which suppressed water intake. It was concluded that the ability of the drugs to induce taste aversion was not related to their unconditioned, hypodipsic effects. However, it was confirmed that when drugs with different durations of action are compared for anorexic or hypodipsic potency, the outcome can be greatly influenced by the time over which measurements are made.
