Effects of several lipidosis-including drugs upon the area postrema and adjacent medullary nuclei of adult rats. I. Alterations is perikarya and dendrites.

The present study is concerned with the question of whether or not amphiphilic drugs (chloroquine, quinacrine, perhexiline) that fail to induce general lipidosis in the central nervous system (CNS) of adult rats can produce lipidosis in a circumventricular organ (area postrema) not furnished with a blood-brain barrier. Chlorphentermine known to induce general lipidosis in CNS of adult rats served as reference compound. All drugs, when chronically applied in high oral doses, induced significant perikaryal lipidosis in the area postrema. In the adjacent nuclei (nucleus tractus solitarii, nucleus dorsalis nervi vagi, nucleus nervi hypoglossi, nucleus gracilis), only chlorphentermine caused generalized lipidosis, whereas the other drugs had either limited or no effects. The present findings strongly suggest that the exemption, of most regions of the CNS of adult rats, from lipidosis induced by chloroquine and others is due to hindered drug distribution across the blood-brain barrier, rather than being due to non-susceptibility of central neurons toward the lipidosis-inducing action of the drugs.

Chronic administration of so-called amphiphilic drugs induces an accumulation of lysosomes of different types containing phospholipids in the inner ear. The inclusions are regularly found in the inner hair cells of the organ of Corti, sometimes in supporting cells, whereas the outer hair cells remain almost unchanged. Several nerve fibres in the area of the inner pillar cells and to a lesser degree in the vestibular labyrinth are swollen and filled with osmiphilic material and vacuoles; sometimes they are degenerated. The preference of the inner hair cells is probably the expression of a specially high turnover of lipids in these cells.

Rapid GC-MS confirmation of urinary amphetamine and methamphetamine as their propylchloroformate derivatives.

Amphetamine and methamphetamine are extracted from 200 microL of urine into an organic solvent containing propylchloroformate. The amines immediately react to form propylcarbamate derivatives. The aqueous phase is removed, and a portion of the organic phase is analyzed by gas chromatography-mass spectrometry. Either the deuterated analogues or N-propylamphetamine internal standards give acceptable results. For day-to-day precision, coefficients of variation in the range of 6-10% are found. The method is linear to 10,000 ng/mL. Limits of quantitation and detection are 50 and 5 ng/mL, respectively, when using N-propylamphetamine as the internal standard. Extracts are stable at room temperature for at least 60 days. There is no interference from other amphetamine drugs.
