The effect of seven vasopressors of halothane MAC in dogs.

We evaluated the effects of ephedrine, mephentermine, metaraminol, phenylephrine, methoxamine, noradrenaline and adrenaline on halothane anaesthetic requirement (MAC) in dogs. MAC increased significantly only during ephedrine infusion (50%) although significance was approached with mephentermine (21%). Phenylephrine, metaraminol, methoxamine, noradrenaline and adrenaline did not change MAC. Arterial pressure was increased 50-100% with all agents. Our results support the hypothesis that anaesthetic requirement may be related, in part, to release of brain noradrenaline.

Myopathy in rats treated with chlorphentermine or iprindole.

Muscular lesions were induced in rats by prolonged administration of chlorphentermine and iprindole. The alterations consisted of longitudinal fibre splitting, fibre degeneration and necrosis, and of formation of dense cytoplasmic inclusions and large cytoplasmic vacuoles. The soleus muscle was more severely affected than were extensor digitorum longus (EDL), gastrocnemius, and lumbrical muscles. This myopathy closely resembles that induced by chloroquine. The pathogenesis of the muscular lesions, and causal relationship between myotoxic and lipidosis-inducing effects of the drugs under study remain to be elucidated.

Effects of prolonged administration of chlorphentermine on the rat lung.

Prolonged administration of the anorexigen chlorphentermine hydrochloride to young male Wistar albino rats for up to one year failed to induce in them either right ventricular hypertrophy or hypertensive pulmonary vascular disease. There was, however, the development of a pronounced pulmonary histiocytosis. Studies by light and electron microscopy showed that these histiocytes disintegrated to liberate their lamellar inclusions into the alveolar spaces, producing a picture reminiscent of alveolar proteinosis. Fibrosing alveolitis occurred in only one animal and was thought to be related to inflammatory changes rather than as a development of the pulmonary histiocytosis. Attention is drawn to the fact that thick-walled large pulmonary arteries are entirely normal in rats and should not be misinterpreted as evidence of hypertensive pulmonary vascular disease in test animals.
